Is there a legal definition of publication? Would a work of art that was created for display in public and is displayed in public like the stained glass of a church for example be considered published?

The legal definition of "publication" is found in 17 USC 101.

As to whether you hypo would be "published," it would depend on the context.

A work is considered published when it is sold or relinquished to a third party for sale, or relinquished for rental, lease or lending.

In your hypo, publication would occur when it was either sold to the church, donated to the church or given to a third party for sale to the church.
