If you receive a one time lump settlement, it very likely will not affect your U.I benefits. You continue to certify and you report any work you did for wages and how much you made during the week you are certifying for.
A large one time cash settlement is not this, and is not reported as wages. Unemployment insurance is NOT an income based program. As long as you are able, available and actively seeking work, you remain eligible, even if you come into a big bunch of money.

That said, it would not be a bad idea to call someone in the unemployment insurance technical support system and discuss this with them, simply to clear it with the state of PA. (717) 783-3140 is the PA information system number.
