Defamation at the workplace?

This story is kinda long, so this is the short version.
About two and a half years ago, at my interview, the supervisor told me that after a year, I'd be able to move to a different department. Move along to about a year ago, we had our company review. At this time, my boss told me that our department was switching formats, and asked if I'd want to move into a more desireable position (the position promosed to me a year and a half earlier). Of course I said yes, and took the necessary route to finding how to transfer. I submitted my resume to the needed person, and waited. A month later, I talked to him, and he said my boss was reviewing it. This continued for 3 months, till I went to my boss. He told me that I didn't have the necessary education. So I went to a department head of the position I wanted, and asked him. He told me that I'd have enough time with the company to transfer, and he'd have to talk it over with his boss. A month later, a job captain from the department I want to work in, asks for my resume. I wait approximately 3 months, and receive an email saying that they are currently looking for people, but only with 4 or 5 year degrees (I have an associates).

That's the background information.
About the same time I received the email, I hear from my manager that someone (99% sure it's my boss) told a department head that I "have a bad attitude", which is why I think I received the email refusing my transfer. Now since this does make my reputation look bad within the company, would this be a possible case for slander/defamation? Also, this comment must have spread through the company, because people that have no business in this issue have said things to me regarding this statement. Now even though I do not have proof which person said this, my manager does know, but refused to tell me, for fear of her own job. I am pretty certain this statement has kept, and will keep, me from advancing through my career with this company, as my boss has a lot of power here.

And I can not leave the company because I am not in a financial position to do so, and there are no available career opportunities in my area, regarding my field of interest/education/experience.
