Would that be an issue if a Green Card holder (through EB2) has self-employed or unemployed history during a couple of years before he/she applied for a US citizenship?

A green card holder is not tied to any employer like a H-1B worker. It is expected that you worked for your sponsoring employer for a period of time after receiving your LPR; Six months is the period of time most frequently mentioned. If you have concerns, you should speak with an attorney before filing forms for citizenship.
