I am suing an ex-coworker for slander. She wrote and signed a false statement, which in turn got me fired. I can't get a copy of this statement, because I did not sign it. Plus, the manager states that the statement is not part of my personnel file, that it is part of his file on me. Is there any way of getting this statement for court?

First, can you somehow get the manager to put in writing that the statement is not part of your personnel file but part of his file on you? [What is the difference?]

I am not a lawyer, this is just what I would do.

In the preparation of your lawsuit, you have the right to 'discovery'. This means that you have a method (suboena) to force the other party to turn over specific records that are needed for your trial preparation.

Contact your court and ask them for the procedures for filing a subpoena for the records that the business has that will help your case.
